Does anyone know if there is a trail/hiking route from Melvin Lake up to Twin Lakes?

No a trail per se, but you can get there along the ridge above Barkley valley.

It's basically a meadow walk from Melvin Lake to Twin Lakes, with a little scree thrown in for good measure.
